Mechanisms involved in the resistance of Mycobacterium tuberculosis to para-aminosalicylic acid.

چکیده
That para-aminobenzoic acid antagonizes competitively the inhibitory effect of paraaminosalicylic acid on the growth of Mycobacterium tuberculosis is well known. In a previous communication (Hedgecock, 1956) it was reported that inhibition of M. tuberculosis (strain H37Rv) by para-aminosalicylic acid was reversed noncompetitively by methionine and biotin when the concentration of the inhibitor did not exceed 0.5 ,ug per ml. At higher concentrations of para-aminosalicylic acid, reversal of inhibition occurred in the presence of methionine and biotin following the addition of certain fatty acids or purines as well as specified amino acids. In the present paper competitive and noncompetitive reversal of para-aminosalicylic acidinhibition was investigated in susceptible and resistant strains of M. tuberculosis which had been isolated from patients and also in resistant strains selected in vitro. Studies of the organisms were made with respect to the mechanisms involved in the development of resistance to paraaminosalicylic acid.

The emergence of Mycobacterium tuberculosis resistant to first-line antibiotics has renewed interest in second-line antitubercular agents. Here, we aimed to extend our understanding of the mechanisms underlying para-aminosalicylic acid (PAS) resistance by analysis of six genes of the folate metabolic pathway and biosynthesis of thymine nucleotides (thyA, dfrA, folC, folP1, folP2, and thyX) and ...

para-Aminosalicylic acid (PAS) entered clinical use in 1946 as the second exclusive drug for the treatment of tuberculosis (TB). While PAS was initially a first-line TB drug, the introduction of more potent antitubercular agents relegated PAS to the second-line tier of agents used for the treatment of drug-resistant Mycobacterium tuberculosis infections. Despite the long history of PAS usage, a...
متن کاملThe Promoter of Rv0560c Is Induced by Salicylate and Structurally-Related Compounds in Mycobacterium tuberculosis
Mycobacterium tuberculosis, the causative agent of tuberculosis (TB), is a major global health threat. During infection, bacteria are believed to encounter adverse conditions such as iron depletion. Mycobacteria synthesize iron-sequestering mycobactins, which are essential for survival in the host, via the intermediate salicylate. Salicylate is a ubiquitous compound which is known to induce a m...

Single nucleotide polymorphisms (SNPs) involved in the development of resistance represent powerful markers for the rapid detection of first- and second-line resistance in clinical Mycobacterium tuberculosis complex (MTBC) isolates. However, the association between particular mutations and phenotypic resistance is not always clear-cut, and phylogenetic SNPs have been misclassified as resistance...
متن کاملEvaluation of Gene Mutations Involved in Drug Resistance in Mycobacterium Tuberculosis Strains Derived from Tuberculosis Patients in Mazandaran, Iran, 2013
Drug resistance (especially multiple drug resistance) in Mycobacterium tuberculosis makes global concerns in treatment and control of tuberculosis. Rapid diagnosis of drug resistant strains of the bacteria has vital importance in the prognosis of the disease. The aim of this study was to identify the mutations responsible for drug resistance in Mycobacterium tuberculosis strains derived from pa...
